Why Use This Instead of Skim Milk?

Skim milk contains proteins such as casein, which can cause cross-reactivity with antibodies and lead to nonspecific signals.
Protein-free blocking solutions help reduce these issues and provide cleaner results.
If you are experiencing problems with high background or detection of nonspecific bands, it is time to try a protein-free blocking solution.

Is it possible to use this instead of BSA for phospho-form antibodies?

Commonly used blocking agents include skim milk and BSA.
Skim milk contains approximately 3.3 g of protein per 100 mL, about 80% of which is casein. Casein is a naturally phosphorylated protein, which can cause high background, especially in experiments using phospho-specific antibodies. Therefore, caution is required when using it.
For this reason, BSA (Bovine Serum Albumin) is often used instead of skim milk, as it contains very few phosphorylated residues structurally. However, BSA may also contain trace amounts of phosphorylated or glycosylated impurities from the manufacturing process, and as a biologically derived protein, lot-to-lot variability may affect experimental results.

Therefore, in experiments using various antibodies such as phospho-form antibodies, glycosylation antibodies, and total protein antibodies, it is more appropriate to use a PROTEIN-FREE blocking agent that minimizes nonspecific binding and reduces background.
